Xenon Pharmaceuticals Shares Sink – Here’s Why

Xenon Pharmaceuticals Inc. (NASDAQ: XENE) shares plunged over 19% in after-hours trading Thursday, September 17, 2026, after the biopharmaceutical company paired a milestone regulatory filing with troubling safety news from its psychiatry drug program.

The Vancouver- and Boston-based company announced it had submitted a New Drug Application (NDA) to the U.S. Food and Drug Administration for azetukalner, its lead drug candidate, as a treatment for focal seizures in epilepsy patients.

The filing was based on positive results from the Phase 2b X-TOLE and Phase 3 X-TOLE2 studies, in which all four tested doses showed statistically significant reductions in monthly seizure frequency versus placebo, with a generally well-tolerated safety profile across more than 1,500 patient-years of data.

However, that good news was overshadowed by Xenon’s disclosure that it has voluntarily paused enrollment of new patients in its ongoing psychiatry studies evaluating azetukalner for major depressive disorder (MDD) and bipolar depression. The move follows an analysis of neuropsychiatric adverse events that had not previously appeared in the earlier Phase 2 X-NOVA study in MDD.

While Xenon said the rate and severity of these events remain consistent with the drug’s known mechanism, the company is now working with its Data Safety Monitoring Board and evaluating potential dosing changes to improve tolerability. Patients already enrolled will remain on their studies.

Enrollment in the pivotal X-NOVA2 MDD trial has reached roughly 360 of an initial 450-patient target — about 80% — which Xenon says is still sufficiently powered for meaningful results. Topline data, previously anticipated sooner, is now expected in the first quarter of 2027, a delay investors appear to be punishing given the setback to Xenon’s broader psychiatric expansion ambitions beyond epilepsy.
